11.6. Methylation dynamics and expression of individual genes
Targeted analyses of selected genes can be
conducted using 454 sequencing of amplified gene fragments from bisulfite-converted
DNAs. For a small-scale testing the amplicons can be cloned into a plasmid,
cloned and sequenced using via Sanger dideoxy sequencing (Foret et al., 2009). Both approaches can be
used to validate genome-wide methylation data, but 454 sequencing allows for a
much higher coverage, as shown in section 8.5.
